Her screen debut came in the 1982 TV movie Parole. From 1986 to 1987 Patricia had a recurring role as Joanne on the television series St. Elsewhere. One of her best known roles came starring as Nancy Krieger Weston in the TV series thirtysomething from 1987 to 1991. In 1996 Wettig starred as Judge Justine Parkes in the series Courthouse. From 1998 to 1999 she starred as Eleanor Riggs in the TV series L.A. Doctors. From 2002 to 2004 Wettig starred as Dr. Judy Barnett in the TV series Alias. She starred as Caroline Reynolds in the series Prison Break from 2005 to 2007. From 2006 to 2011 she starred as Holly Harper in the TV series Brothers & Sisters. Patricia appeared in 85 episodes of Brothers & Sisters between 2006 and 2011.

Wettig has starred in the movies Guilty by Suspicion, City Slickers, Me and Veronica, City Slickers: The Legend of Curly's Gold, Bongwater, and Nightmare in Big Sky Country. Patricia won a Golden Globe in 1991 as well as three Primetime Emmy Awards for thirtysomething.
